Two different mechanisms are considered the main causes of aging.

One of these mechanisms (concept ROS) are the DNA damage caused by the accumulation of reactive oxygen species (ROS), the by-products of oxidative phosphorylation. The second mechanism (concept TOR) is the constitutive stimulation of mitogen-and nutrient sensing mTOR/S6.

Have been used the flow and the laser scanning cytometry to measure the level of DNA damage constitutive / ros and signaling mTOR/S6- in single cells.


Potential gero-suppressive agents such as rapamycin, metformin, 2-deoxyglucose, berberine, resveratrol, vitamin D3 and aspirin all have reduced the level of DNA damage constitutive.All have also decreased the level of intracellular ROS and mitochondrial transmembrane potential ΔΨm, mitochondrial marker of excitement as well as reduced phosphorylation of mTOR, RP-S6 and 4EBP1.

Although the primary objective of each of these agents is different, the data agree in indicating that produce a drop of mTOR/S6K coupled with a decrease in oxidative phosphorylation, which leads to the reduction of ROS and therefore the oxidative damage of DNA.

The rate of decrease induced by these agents may slow the cell hypertrophy and alleviate other characteristics of cell aging / senescence.
